Microrheology Formulation

Particle tracking microrheology (PTμКR) is a technique with the potential to contribute significantly to our ability to monitor protein unfolding and aggregation and help improve our understanding of parameters such as the manufacturing design space (manufacturability) and the injectability of protein formulations. Microrheology is a well-established technique in the fields of colloids and soft condensed matter, but has only recently been applied to protein formulations.

Microrheology measures the mobility of nanoparticles moving through a solution due to Brownian motion. This mobility is affected by mechanical properties of the surrounding solution. Since proteins change their size when they unfold and aggregate formation will affect the mobility of the tracer particles, microrheology should be an ultrasensitive probe of the state of proteins in a formulation...
