Soft gelatine capsules often improve the performance of drugs insoluble in water or gastric juices. Certain drugs such as neutral compounds or organic acids are insoluble in water and gastrointestinal fluids and, if presented as tablets or hard shell capsules, the absorption rate is low and bioavailability correspondingly poor. In these cases, several methods can be employed to minimise the problem (see Figure 1). For example, neutral compounds may be formulated as a fine suspension, using powders or dispersible tablets, although such presentations are often not the ideal form for marketing purposes.
Alternatively, combination with surfactants or polymers can produce a solid solution that will dissolve rapidly but which may be chemically unstable or waxy, making it difficult to process.
